Antiproliferative activity of daidzein and genistein may be related to ERα/c-erbB-2 expression in human breast cancer cells

In this study, we investigated the antiproliferative activity of the isoflavones daidzein and genistein in three breast cancer cell lines with different patterns of estrogen receptor (ER) and cerbB2 protein expression (ERαpositive MCF7 cells, cerbB2positive SKBR3 cells and ERα/cerbB2positive ZR751). After treatment at various concentrations (1200 µM for 72 h), the effect of daidzein and genistein on the proliferation of different cell types varied; these effects were found to be associated with ERα and cerbB2 expression. Daidzein and genistein exhibited biphasic effects (stimulatory or inhibitory) on proliferation and ERα expression in MCF7 cells. Although 1 µM daidzein significantly stimulated cell growth, ERα expression was unaffected. However, genistein showed marked increases in proliferation and ERα expression after exposure to <10 µM genistein. Notably, the inhibition of cell proliferation by 200 µM genistein was greater compared to that by daidzein at the same concentration. Daidzein and genistein significantly inhibited proliferation of SKBR3 and ZR751 cells in a dosedependent manner. In addition, ERα and cerbB2 expression was reduced by daidzein and genistein in both SKBR3 and ZR751 cells in a dosedependent manner. However, the effect of genistein was greater compared to that of daidzein. In conclusion, the isoflavones daidzein and genistein showed antibreast cancer activity, which was associated with expression of the ERα and cerbB2 receptors.

在此研究中,我们研究了异黄酮苷元和金雀异黄素在三种雌激素受体 (ER) cerbB2 蛋白表达模式不同的乳腺癌细胞(ERα阳性 MCF7 细胞,cerbB2阳性 SKBR3 细胞和 ERα/cerbB2阳性 ZR751)中的抗增殖活性。在不同浓度 (1200 µM72 h) 治疗后,大豆苷元和金雀异黄素对不同类型细胞增殖的影响各不相同;发现这些影响与 ERα cerbB2 的表达有关。大豆苷元和金雀异黄素对MCF7 细胞的增殖和ERα 表达表现出双向性作用 (刺激或抑制)。虽然 1 µM 大豆黄酮显著刺激细胞生长,ERα 的表达不受影响。然而,当接触< 10 µM 金雀异黄素以后,金雀异黄素在细胞增殖和ERα 表达表现出明显的提升。值得注意的是, 200 µM 金雀异黄素对细胞增殖的抑制作用比相同浓度下的大豆黄酮更大。大豆苷元和金雀异黄素以剂量依赖的方式明显抑制了SKBR3 ZR751 细胞的增殖。此外,ERα cerbB2 表达被大豆苷元和金雀异黄素在 SKBR3 ZR751 细胞中以剂量依赖的方式下调。然而,金雀异黄素的影响相比大豆黄酮较大。总之,异黄酮大豆苷元和金雀异黄素表现出抗乳腺癌活性,并且与 ERα cerbB2 受体的表达有关。
